Effect of PD-1 pathway in immune cells of imiquimod-induced psoriasic mouse model

ABSTRACT  Objective: To explore the role of programmed death 1(PD-1) and programmed death ligand 1(PD-L1) pathway in immune cells of psoriasis mice.Methods:Sixteen BALB/c mice were randomly divided into the model group(n=8) and the control group(n=8).Imiquimod cream was smeared on the mice in the model group to induce psoriasis.After the psoriasic mouse model was established,the levels of PD-1 and PD-L1 in immune cells of spleen and peripheral blood of mice both in the model group and the control group were detected by flow cytometry.Results:The spleens of the mice in the model group were larger than those in the control group.In the increased spleen cells,the expression levels of total PD-1 and PD-L1 all increased.The expression of PD-1 in T helper cells and the expression of PD-L1 in dendritic cells (DC) and plasmacytoid dendritic cells (pDC) increased.In peripheral blood,the expression of PD-1 and PD-L1 also increased.The expression of PD-L1 in DC increased,however,the expression of PD-1 in T helper cells and PD-L1 in pDC all down-regulated.Conclusion:The down-regulation of PD-1 in T helper cells and PD-L1 in pDC in peripheral blood may induce the dysregulation of immune cells in psoriasis mice,thereby causing or aggravating progress of psoriasis.
