Quantitative determination of fosfomycin calcium and its related substances by ion-pair HPLC method

ABSTRACT  Objective:To establish a practical ion-pair HPLC method for the determination of contents of fosfomycin calcium and its related substances. Methods: The column was Synergi 4μ Fusion-RP 80A (250 mm×4.6 mm, 5 μm) and 15 mmol/L octylamine solution (adjusted to pH 4.8 with glacial acetic acid) - acetonitrile (94∶6) was mobile phase at the flow rate of 1.5 mL/min.The column temperature was 35 ℃. Evaporation light scattering detection (ELSD) was used as the detection method and nitrogen was used as the nebulizing gas with the pressure of 1.6 L/min.The evaporation temperature was 50 ℃. Results:The linearity of fosfomycin calcium concentration was good within the range of 0.485-1.940 mg/mL and 0.05-0.40 mg/mL(r = 0.999 8 and 0.999 6,respectively). The RSD of fosfomycin calcium and 1,2-dihydroxypropyl phosphonate calcium (according to British pharmacopoeia) were 0.5% and 1.6%,respectively. The precision and accuracy were good. The test solution was stable within 4 hours. Conclusion:This method is simple, accurate and rapid. It is suitable for quality control of fosfomycin calcium and quantitative determination of its related substances.
