Effect of lycopene on the vascular endothelial function and antioxidative activity in hyperhomocysteinemic rats

ABSTRACT  Objective: To investigate the mechanism of action lycopene in preventing atherogenesis induced by hyperhomocysteinemia(HHcy) in rats.Methods:Fifty male SD rats were randomly divided into 5 groups: normal control group(NC),which was fed with Association of Official Analytical Chemists(AOAC) diet, and 4 HHcy groups,which were fed with AOAC diet plus 3% L-methionine and were daily supplemented with 0(HC),10(HL1),15(HL2),20 mg/kg(HL3) of lycopene for 12 weeks, respectively.Serum levels of homocysteine(Hcy),nitric oxide(NO),nitric oxide synthase(NOS),endothelin-1(ET-1),malondialdehyde(MDA),hydroxy radical(OH·),hydrogen peroxide(H2O2),total antioxidative capability(T-AOC),oxidized low density lipoprotein (OX-LDL),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),catalase(CAT) and lactate dehydrogenase(LDH) were determined,and the pathological observation of abdominal aorta was done.Results:Moderate hyperhomocysteinemia was induced in HHcy groups.Serum level of NO was lower and ET-1 was higher in HC group than in NC,HL2 and HL3 groups(P<0.01).There was no significant difference in serum level of NOS in these 5 groups.There were break and deposition of fat granutes on aortic tunica intima in HC and HL1 groups,which were not found in NC,HL2 and HL3 groups. Serum levels of MDA and H2O2 were higher in HC and HL1 groups than in NC group (P<0.01),and were decreased significantly in HL2 and HL3 groups compared with HC group (P<0.05). Serum level of OH· was lower in HL2 and HL3 groups than in HC group (P<0.01). In contrast to NC group, the serum level of T-AOC showed a significant reduction and that of OX-LDL rose sharply (P<0.01) in HC group. With the increase in dose of lycopene, the serum level of T-AOC in different groups of HHcy rats increased and that of OX-LDL decreased(P<0.01). Serum activities of SOD and GSH-Px were lower in HC group than in NC group (P<0.01), and were higher in HL1,HL2 and HL3 groups compared with HC group (P<0.05). There was no significant difference in serum activities of CAT in these 5 groups. Conclusion: This study indicated that middle and high dose of lycopene could prevent atherogenesis induced by HHcy, which is probably associated with the action of lycopene in directly quenching reactive oxygen species, enhancing SOD and GSH-Px activities, and accelerating the inactivation of reactive oxygen species.
