Effects of supplement with glucose on the model rats for the investigation of extrahepatic metabolism

ABSTRACT  Objective: To explore the effects of supplement with glucose on the survival time and blood biochemical indices of the model rats for the investigation of extrahepatic metabolism. Methods: Rat model for assessment of extrahepatic metabolism was established by ligating the hepatic artery and clamping the portal vein. Sixteen model SD rats were randomly divided into two groups, modelⅠgroup and modelⅡgroup, of 8 rats each. The rats in modelⅠ group were injected intragastrically with 25% glucose 3 mL, and those in model Ⅱ group were injected with 3 mL water after the operation. Their survival time was observed. Additional 29 SD rats were randomly divided into normal control group (n=6) and model group(n=23). The serum concentrations of glucose, bilirubin, creatinine, urea nitrogen,uric acid, K+, Na+ and Cl-were determined at 0.5 (n=3), 1 (n=3), 2 (n=12) and 4 h (n=5) after the operation, respectively. Of the 12 rats whose blood samples were taken at 2 h, 6 were injected intragastrically with 25% glucose 3 mL after operation and 6 remainders were injected with water 3 mL.Results:The survival time of model rats was (6.7±1.6) h, and it was prolonged to (14.5±3.2) h by the supplement with glucose(P< 0.01).The serum concentration of glucose decreased by 52.3% and 95.0% at 0.5 and 4 h after the operation in the model rats, while serum bilirubin increased by 17.2-fold. The serum concentrations of creatinine, urea nitrogen and K+ began to rise at 1 h after the operation, which was significantly inhibited in the rats supplemented with glucose. Conclusion:The principal pathophysiologic changes of model rats are characterized by hypoglycemia, hyperbilirubinemia and renal insufficiency. The supplement with glucose after the operation can prolong the survival time of model rats and improve the renal function.
