Formulation optimization and in vitro percutaneous penetration of compound phellodendri Chinese cortex fluid

ABSTRACT  Objective: To study the effects of improved formula of compound phellodendri Chinese cortex fluid on in vitro percutaneous penetration of the drug,so as to provide evidence for formula optimization and production quality control. Methods: With the help of double-chamber penetration unit,optimal percutaneous penetration fluid was screened by using the shaven skin of the rat abdomen as the experimental barrier,and the volume of cumulative penetration of berberine hydrochloriade within the pre-set time was measured as the evaluation indicator by HPLC method. At the same time,percutaneous penetration rates of 3 improved formula preparations were investigated by adding 1% borneol,1% menthanol and 0.5% borneol plus 0.5% menthanol. Results: (1) The linear relationship was good in the range from 0.003 72 μg to 0.023 8 μg for berberine hydrochloride (R2=0.999 8). (2) When 0.9% sodium chloride (normal saline,NS),NS of 20% ethanol,NS of 40% ethanol,20% polyethylene glycol (PEG) were used as the accepted liquid,the cumulative penetration valumes of berberine hydrocholoride in 24 hours were 10.304,7.997,6.551 and 6.074 μg, respectively. Therefore,NS was used as the accepted liquid.(3) The cumulative penetration of berberine hydrochloride of the improved prescriptions in 24 hours by adding 1% borneol or 1% menthanol or 0.5% borneol plus 0.5% menthanol were 14.014,13.291 and 17.895 μg,respectively,which was better than that of original formula (10.304 μg). Conclusion: The percutaneous penetration of the compound phellodendri Chinese cortex fluid of the improved formula was significantly superior to that of the primary formula.
