Study on the preparation and in vitro transdermal characteristics of ketoconazole ethosome gel

ABSTRACT  Objective:To prepare the ketoconazole ethosome gel and study its in vitro transdermal characteristics.Methods:The ethosome and liposome were prepared by ethanol injection method and the gels were prepared using mixing method. The drug transdermal characteristics across the excised rat skin and drug accumulation in skin were evaluated by Franz diffusion cell method and skin homogenation method,respectively.The concentration of ketoconazole was determined by HPLC method.Results: The transdermal rate of ketoconazole in ethosome gel was 1.41 and 1.07 times faster than those in the regular gel and liposome gel,respectively.While the accumulation of ketoconazole in skin was 12.77 and 2.08 times higher than those in the regular gel and liposome gel,respectively.Conclusion: The ketoconazole ethosome gel was an excellent local cutaneous agent.
