Cardioprotective effects of total flavonoids extracted from Apocynum venetum L.on ischemia/reperfusion-induced myocardial injury and its mechanism

ABSTRACT  Objective: To evaluate the cardioprotective effects of total flavonoids extracted from Apocynum venetum L.(ALTF) on ischemia/reperfusion (I/R) injury using an isolated Langendorff rat heart model.Methods: A model of isolated myocardial I/R injury in rats was established using Langendorff isolated heart perfusion system.The isolated rat hearts were divided into the control,the I/R model,and the ALTF 1,5,25 μg/ml groups.The homodynamic parameters of cardiac arrest and reperfusion in vitro,including the left ventricular developed pressure (LVDP) and the maximum up/down rate of the left ventricular pressure (±dp/dtmax) were recorded by BL-420F instrument biology polygraph.The activity of superoxide dismutase(SOD) and the levels of malondialdehyde (MDA),tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) were measured in the myocardium.Cardiomyocyte apoptosis analysis was performed by TUNEL method.Results: Compared with the control group,the LVDP and ±dp/dtmax were remarkably decreased,the levels of MDA,TNF-α and IL-6 were significantly increased,and the activity of SOD was significantly decreased in the I/R model group.Obvious cardiomyocyte apoptosis was shown in the I/R model group.Compared with the I/R model group,the LVDP and ±dp/dtmax were increased significantly,the activity of SOD in myocardial tissue was significantly increased,and the contents of MDA,TNF-α and IL-6 were significantly decreased in the ALTF 5 and 25 μg/ml groups.The myocardial cell apoptosis rate was lower in the ALTF 5 and 25 μg/ml groups as compared with that in the I/R group.Conclusion:ALTF could produce significant protective effect on myocardial injury,the mechanism of which might be closely associated with its anti-oxidant,anti-apoptosis and anti-inflammatory activity.
