Effects of alternol on proliferation and melanin synthesis of human melanoma cells

ABSTRACT  Objective:To study the effects of alternol on the proliferation and melanin synthesis of human A375 melanoma cells.Methods:A375 melanoma cells were incubated with different concentrations(0,0.3,0.6,1.5,3.0 and 4.0 μg/ml) of alternol for 24 and 48 h.Subsequently,the inhibition rate of cell proliferation was detected by tetrazolium salt colorimetric assay(MTT).Survival rate of A375 cells treated with alternol was determined by trypan blue exclusion staining test.The morphological changes of A375 cells were detected by Giemsa staining.Sodium hydroxy alkaline lysis method and dopa oxidation method were used to measure the content of melanin and activity of tyrosinase,respectively.Results:Alternol inhibited the proliferation of A375 cells significantly after incubation for 24 or 48 h in a time-dependent and concentration-dependent manner.After treatment with alternol,obvious morphological changes of A375 cells were observed,i.e.dentric projections.Tyrosinase activities and melanin content were increased obviously with the increase of alternol concentration.Conclusion:Alternol could surpress the cell proliferation,change cellular morphology,increase melanin level and tyrosinase activity of A375 cells in a concentration-dependent manner.
