Enantiomorph of cetirizine hydrochloride by HPLC method with cellulose derivative chiral stationary phase

ABSTRACT  Objective: To establish a chiral stationary phase HPLC method for the enantiomorph of cetirizine.Methods:CHIRALCEL OD-H column (250 mm×4.6 mm,5 μm) was used. Effects of the component proportion of mobile phase,proportion of acid additive and flow rate on enantiomorph were investigated.Results:The optimized chromatographic conditions were as follows:the mobile phase was hexane∶ethanol∶ trifluoroacetic acid(80∶20∶0.05,V/V),with a flow rate of 0.8 ml/min and the detection wavelength of 230 nm. The column temperature was set at 25 ℃.The cetirizine enantiomers could be well separated with a resolution factor of 1.64 and the resolution time for d- and l-cetirizine were 9.6 min and 12.0 min,respectively.Conclusion:This method is simple to operate and could be used for the separation and quality control of cetirizine enantiomers.
