Triptolide enhances in vitro apoptosis of human gallbladder cancer NOZ cells induced by fluorouracil

ABSTRACT  Objective: To investigate the effects of triptolide and fluorouracil(5-FU) on the proliferation and apoptosis of gallbladder cancer NOZ cells in vitro.Methods: NOZ cells were cultured in the William’s Medium E containing 10% fetal bovine serum in 5% CO2 at 37 ℃.The proliferation and apoptosis effects of triptolide,fluorouracil and combined use of the two drugs on NOZ cells were determined by CCK-8 assay,Annexin V-FITC/PI bistaining and cell cycle analysis.Results: Both triptolide and fluorouracil alone,and combined use of the two drugs all inhibited the proliferation and induced apoptosis of NOZ cells.The CCK-8 assay showed that the IC50 of both the triptolide and fluorouracil group and the combination group were respectively 15.6,11.2 and 6.85 μmol/L,48 h after treatment.The Annexin V-FITC/PI bistaining showed that the apoptosis rate of NOZ cells in the combination group was significantly higher than those in the single medication groups(P<0.05).The cell cycle analysis showed that the proportion of NOZ cells at G50 stage in the combination group was significantly higher than those in the single medication groups(P<0.05).Conclusion: Triptolide could significantly enhance the in vitro inhibitory and apoptosis effects of fluorouracil on human gallbladder cancer NOZ cells.
