Metabonomic study on the anti-osteoporostic effect of active ingredients of Epimedium brevicornu and Curculigo orchioides

ABSTRACT  Objective: To observe the differences of metabolite profiles of urine and serum samples between ovariectomized(OVX) rats and OVX rats treated with total flavonoids(TF) of Epimedium brevicornu,total benzyl benzoate glucosides(TBBG) of Curculigo orchioides and their combination,so as to explore the synergistic effects of TF and TBBG for treating osteoporosis.Methods: 36 female SD rats were randomly divided into 6 groups, the sham operation group,the model group,the positive control group,the TF treatment group,the TBBG treatment group and the combination treatment group, each consisting of 6 mice. 05% CMC-Na was used as a solvent to prepare drugs with a volume of 10 ml/kg.Rats in the sham operation group and the model group were given 10 ml/kg physiological saline.Rats in the positive control group were given nilestriol at a dosage of 1 mg/kg once a week.Rats in the TF treatment group were given 100 mg/kg TF,rats in the TBBG treatment group were given 50 mg/kg TBBG, and rats in the combination treatment group were given 100 mg/kg TF and 50 mg/kg TBBG.The rats were gavaged with the drugs, once a day,six times a week, for a duration of 12 weeks. Bone mineral density(BMD) and oxidative stress indexes in serum were measured,and serum and urine samples were collected for 1H-NMR analysis.Partial least square method was used to identify and analyze characteristic metabolites,and their metabolic pathways were also speculated.Results: The effective ingredients of E. brevicornu and C. orchioides could all significantly increase BMD in OVX rats,and the compatibility of the two drugs displayed a synergistic effect. 13 metabolites in serum and 16 metabolites in urine were identified by partial least square method.TF combined with TBBG showed significant synergistic effects on the regulation of blood glucose,blood lipids,and levels of taurine and glutamate in urine.Conclusion: The synergistic anti-osteoporosis effect of E. brevicornu and C. orchioides might be associated with the synergistic regulation of glucose,taurine and glutamate metabolism.
