Screening of the extraction methods of Lingxianjiangsuan compound with pharmacodynamics as an index

ABSTRACT  Objective: To study the extracts from Lingxianjiangsuan compound by different extraction methods with pharmacodynamics as an index,so as to screen out the optimal extraction method.Methods: The Wistar rats were gavaged with adenine and potassium oxonate to establish hyperuricemia models,and then upon successful establishment of the model,40 rats were randomly divided into the model group, the Lingxianjiangsuan compound water extraction group,the water extraction and alcohol precipitation group,the alcohol extraction group (10 g crude drugs/kg),the positive controlled allopurinol treatment group(20 mg/kg) and the blank control group.The drugs were administered by gavage once daily for 21 d.The levels of uric acid(UA),urea nitrogen(BUN),serum creatinine(CR) were determined on days 7,14 and 21,respectively. On day 21, the rats were excuted,after blood samples were taken,and viscera were obtained. Then the level of adenosine deaminase(ADA) in liver and kidney weight were measured.Results: Compared with those of the model group,the levels of UA,BUN and CR,kidney weight,level of ADA in liver of the water extraction group were significantly decreased(P<005,P<0.01),while some indicators in the water extraction and alcohol precipitation group and the alcohol extraction group were significantly difference,and some indicators showed a decreasing trend.Conclusion: The water extract of Lingxianjiangsuan compound has better effects on the reduction of UA and alleviation of pathological damage to kidney, as compared with the water extraction and alcohol precipitation and alcohol extraction, and the optimal extraction method of Lingxianjiangsuan compound is water extraction.
