Exploration of glycopeptide biomolecular markers of gastric cancer by liquid chromatography-mass spectrometry

ABSTRACT  Objective: To explore potential N-glycopeptide biomolecular markers in serum of gastric cancer patients.Methods: The N-glycopeptides in the serum samples of patients with gastric cancer and gastritis were quantitatively analyzed by ultra-high performance liquid chromatography coupled with triple quadruple mass spectrometry(UPLC-QqQ-MS/MS).The data were analyzed by independent samples t-test and partial least squares(PLS) methods.Results: Seven N-glycopeptides with significant differences were found among about sixty different N-glycopeptides,namely N4400-IgG1,N5400-IgG1,N4410-IgG1,N3500-IgA,N5400-IgA,N5402-IgA and N5501-J.The results showed that there was a certain separation tendency between patients with gastritis and gastric cancer.Conclusion: Potential N-glycopeptides biomolecular markers for gastric cancer were identified.This method provides powerful technical support for exploring biomarkers of gastric cancer,and is of important clinical significance for early diagnosis of gastric cancer.
