Study on the moisture absorption/moisture retention of yam polysaccharides and its transdermal features in vitro

ABSTRACT  Objective: To investigate the moisture absorption/moisture retention features of yam polysaccharides, as well as its transdermal absorption rate in vitro, so as to provide experimental evidence for its further development and application.Methods: The moisture absorption capacity of yam polysaccharides under the conditions of relative humidity of 43% and 81% were determined,and the moisture retention capacity in the dry environment created by dry micronized silica gel was also determined.The steady-state transdermal penetration rate,24-h cumulative permeability and skin retention rate of yam polysaccharides were determined by using a modified Franz diffusion cell.Results: Under the conditions with relative humidity of 43% and 81%,the moisture absorption rates of yam polysaccharides at 24 h were 37.6% and 48.4%,and at 48 h were 40.2% and 55.3%,respectively,which were lower than sodium alginate and higher than glycerol.In the dry environment created by dry micronized silica gel, the moisture retention rates of yam polysaccharides at 24 h and 48 h were 24.8% and 16.5%,respectively,which were higher than sodium alginate and glycerol.The steady-state transdermal penetration rate of 10% yam polysaccharide solution in the modified Franz diffusion cell was 1.460 μg·cm-2·h-1,the cumulative permeability at 24 h was (1.87±0.31)%,and the skin retention rate was (0.60±0.10)%.Conclusion: The yam polysaccharides have good properties of moisture absorption,moisture retention and transdermal permeability.
